Abstract
Background. Valproic acid (VPA) is a histone deacetylases inhibitor. VPA inhibits cell proliferation, induces apoptosis. VPA has a diuretic effect related to Na-K-Cl cotransporter (NKCC) inhibition in rat kidney. NKCC in somatic cells is responsible for chloride (Cl-) influx into the cell. The intracellular Cl- level is one of the messengers in cell proliferation. Cl- transport in thymocytes was described. NKCC is a marker of apoptosis and proliferation. VPA has been studied for various cancers treatment. The SLC5A8 transporter is a Na+-coupled monocarboxylates transporter (SMCT1). SLC5A8 is a candidate tumor suppressor gene, which expression is related to enhanced histone acetylation and apoptosis in the VPA-treated cells. The aim of the study was to investigate the effect of VPA on thymus weight, its structure, and NKCC, SLC5A8 RNA expressions in thymocytes. Methods. Wistar rats, age 4 to 5 weeks, were investigated in VPA-treated gonad-intact and castrated males and females and in control groups (n=6 in a group). The VPA 300 mg/kg/day treatment duration was 4 weeks in drinking water. The NKCC (a Slc12a2 gene) and SLC5A8 (a Slc5a8 gene) RNA expressions were determined by real-time PCR method; thymus cortical, and medullary areas structural changes were assessed in slides stained with H–E; Hassall‘s corpuscles numbers per 1 mm2 (HCs) were determined immunohistochemically.
