Abstract
Background. Glioblastoma is the most malignant brain tumor in adults and children. Pediatric glioblastoma differs from adult glioblastoma in terms of cancer biology, progression. Sodium dichloroacetate (DCA) is an investigational anticancer preparation. DCA, inhibiting pyruvate dehydrogenase kinases, reverses a glycolytic cancer cells phenotype, increases reactive oxygen species generation, inhibits Na+-K+-2Cl– cotransporter, induces tumor cell apoptosis. The chick embryo chorioallantoic membrane (CAM) tumor xenograft model is an outstandingt system for tumor growth, its treatment efficacy, angiogenesis studies. The aim of the study was to examine the DCA effect on pediatric and adult glioblastoma xenograft (GB) invasion into CAM frequency, the CAM thickness under tumor, and the number of blood vessels in CAM, and to compare the differences between them. Methods. The GB (adult female’s U87 and 13-year-old boy’s PBT24 cell line cell tumors grafted on CAM) were studied. A window was drilled in the shell of the Cobb 500 fertilized chicken egg on the day 3 of embryo development (EDD3). The GB was formed with 1×106 tumor cells and rat tail collagen, formed tumor was grafted onto CAM on EDD7. The study groups were as following: 1) U87-control (non-DCA treated, n=15), treated with 10 mM of DCA (U87-10 mM, n=15), U87-5 mM (n=15); PBT24-control (n=11), PBT24-10 mM (n=10), PBT24-5 mM (n=10).
